is Mailbox automatically set when new domain is added to the O365 tenant?

When we create new O365 tenant and adding custom domain(Example: mycustomsite.com). Will all the users in the domain added to the tenant automatically? can users able to access the outlook without license?

    No, each account must be licensed with an Exchange Online license to get and logon to a mailbox.
